High yields across diverse donors

The total DNA yield is a function of WBC count. The HMW DNA extraction app consistently delivers >1 µg of high-quality HMW DNA from samples with >6 million cells/mL, providing sufficient input for long-read sequencing. DNA concentration was measured using Qubit dsDNA HS.

DNA consistently meets fragment length and purity specs for all downstream library preps.

Callisto platform produces HMW DNA with high purity and long fragment length. Purity (Left): A260/280 ratio consistently falls within the 1.7 – 2.2 range (600 samples) indicating no protein contamination. Fragment length (Right): Genomic Quality Number (GQN) represents the percentage of a sample's total DNA that exceeds a specified size threshold, indicating its suitability for long-read sequencing. Callisto platform produces GQN10Kb score of >9 (90% of fragments are above 10 kb) and GQN40Kb >6 (60% of fragments are above 40 kb) .

Achieve industry-leading fragment lengths with minimal hands-on time

Callisto outperforms five widely used HMW DNA extraction kits across multiple chemistries—including magnetic bead, disc, glass bead, column, and PEG-based volume exclusion methods. While yields (normalized yields per microliter) and purity are comparable, Callisto delivers significantly longer DNA fragments with far less hands-on time. Manual methods require 3–6x more hands-on time, making Callisto the superior choice for efficient, high-quality extractions.

High-quality PacBio sequencing using HMW DNA extracted on Callisto

PacBio sequencing was performed on libraries prepared using VoltaPure HMW DNA extraction and PacBio library prep Apps  on Callisto. Key sequencing metrics—including mean read length, N50, and read length distribution meets PacBio sequencing specifications—demonstrates DNA extracted on Callisto is suitable for PacBio sequencing. Similar HiFi read lengths distribution  across replicates, highlighting the reproducibility. Blue pippin was used to remove fragments <10 kb for optimal sequencing.
